Method step description:
- -Identificación de las muestras: Rotular la muestra con marcador no borrable Nombre del lote o código del material vegetal, parcela, municipio. Fecha. Nombre del Recolector.
- -RECOLECTA DE MUESTRAS DE SUELO. Se colectan submuestras de suelo tomadas al azar de un terreno que presente características homogéneas. Para esto se dividirá el terreno en las zonas topograficas que presente, zonas altas, bajas, ondulaciones, inundadas. Se rotularan las muestras en la bolsa y con cinta de enmascarar y se anotará en el cuaderno de registro. Recolección: Una vez delimitado el terreno se colectan muestras aditivas para un total de 25 muestras por hectárea de suelo, de aproximadamente 50 gramos cada una. Se realiza un trazo en zigzag en el área para delimitar el espacio o se recolectó material asociado a un árbol. En el punto de muestreo se debe remover el material vegetal, hojarasca o raíces. Posterior a esto se procede a lavarse las manos con alcohol antiséptico, esterilizar el barreno metálico limpio con alcohol y fuego en el mechero. Una vez se esterilice se entierra el barreno en el punto de muestreo, girándolo como si fuera un destornillador sobre un tornillo. Luego de esto se retira y se deposita la muestra en la bolsa ziploc nueva que ya debe estar rotulada. Se adicionan en una sola bolsa todas las submuestras. Una vez de ha completado la totalidad de las submuestras se mezcla bien hasta homogenizarlas y la muestra se mantiene refrigerada hasta llevarla al laboratorio.
- -RECOLECTA DE FRAGMENTOS DE RAMA Se estudiara el árbol seleccionado buscando ramas jóvenes que no presenten lesiones flores o frutos. Se rotularan las muestras en la bolsa y con cinta de enmascarar y se anotará en el cuaderno de registro. Lavarse las manos con alcohol antiséptico. Se esterilizaran las tijeras en la llama del mechero y se cortará un fragmento de 10 cms de largo de la rama seleccionada. El fragmento será guardado en una bolsa ziploc limpia por muestra, La muestra debe mantenerse refrigerada hasta el momento de análisis en el laboratorio.
- -RECOLECTA DE FRUTOS Se determina la presencia en el árbol seleccionado de un número adecuado de frutos y se registra el estadio de madurez. Se rotularan la bolsa con el borrador indeleble y con cinta de enmascarar y se anotará en el cuaderno de registro. Lavarse las manos con alcohol antiséptico. Se esteriliza las tijeras en la llama del mechero. Se abre la bolsa y se coloca el fruto en su interior sin tocar el fruto. Ir cerrando la bolsa y se corta el fruto, al finalizar se sella totalmente la bolsa. El fruto es mantenido refrigerado hasta el momento de análisis en el laboratorio.
